The evidence is consistent with a correlation between stimulation of HSV-1 infection and abrogation of the sumoylation and stability of PML and Sp100, the disruption of PML NBs, and the inhibition of recruitment of PML to the sites of HSV-1 genomes, thus further strengthening the previous evidence that gave rise to these connections. Nonetheless, a degree of caution regarding this interpretation remains necessary. Both ICP0 (1) and Gam1 (17,–19, 22) affect a wide variety of cellular proteins, and although the correlation with PML NB-related components is high, it is possible that ICP0 and Gam1 employ functionally analogous mechanisms that affect other cellular proteins that have major regulatory roles and that have not been analyzed in this study. In this scenario, PML NB components and sumoylated proteins could act as excellent proxies for these hypothetical regulatory factors. The simplest hypothesis remains, however, that PML NB- and sumoylation-related mechanisms play a major role in regulating HSV-1 infection.
